How to determine the right dose?
Dose finding can be expensive in terms of time, money and, more importantly, our aim should be to minimize the number of patients exposed to either ineffective or dangerous doses. However, we cannot know which doses fall into which category from Day 1! A classical design will lock us into fixed patient numbers so we are increasingly looking to adaptive trial design (ATD) with re-calculation of randomized patient numbers across different dosage groups based on accumulating data, including closing ineffective or dangerous cohorts as soon as possible.
To make these decisions, we need a data set that is current, clean and comprehensive.
